表油菜素内酯(EBR)是油菜素类固醇,类固醇衍生植物生长调节剂家族的生物活性化合物。将细胞与各种剂量(0-100μM)的表油菜素内酯一起温育24或48小时,并通过MTT测定法测定细胞活力。与LNCaP和DU145前列腺癌细胞中的未处理样品相比,表油菜素内酯以剂量和时间依赖性方式诱导细胞活力丧失。与DU145细胞相比,增加浓度的表油菜素内酯对LNCaP细胞活力丧失更有效,这表明雄**依赖性细胞对表油菜素内酯比对雄**非依赖性前列腺癌细胞更敏感。在进一步的实验中,选择25μM的表油菜素内酯,因为其对两种细胞系具有中等的细胞毒性作用。通过在96小时内计数细胞数来检查表油菜素内酯治-疗对细胞增殖的影响。在LNCaP中观察到比DU145细胞更高和更早的细胞增殖抑制[1]。
